Abstract: 

The Hyoscyamus genus has always been regarded as an important medical plant and its therapeutic effects are attributed to its contained different tropan alkaloids. Utilizing genetic engineering for production of plants belonging to this genus with more valuable pharmaceutical properties without consuming much time and cost is one of the most regarded points in this issue. Somatic embryogenesis and gathering knowledge about it is a great help. For this perpose we attempted to perform successful somatic embryogenesis, and to get more knowledge about its mechanism via assessing ENZYMES in different stages of embryo formation. In this study, leaves were cultured on MS medium containing BAP (1.5 mgL-1) and IAA (0.2mgL-1) lead to a successful embryogenesis. The results show that highest activity of superoxide dismutase and peroxidase had the highest activity in primary embryo-holding callus, catalase and ascorbate peroxidase had the highest activity in embryogenic callus but polyphenol oxidase had the highest activity in callus. In different stages of embryo formation from leaf explants of H. niger showed that changes of activities of these ENZYMES in various stages of embryo formation. The electrophoresis patterns of iso-ENZYMES of superoxide dismutase, peroxidase, catalase, ascorbate peroxidase, polyphenol oxidase and auxin oxidase showed the presence of 7, 6, 1, 5, 6, 7, and 2 isosymes in these ENZYMES respectively. The molecular mass of the isozymes were determined and up-regulation, down-regulation, and modulation of the isosymal bands were assessed. The main as well as concervative isozymes involved in embryogenesis were also identified.

Abstract: 

It has been accepted that oxidative damage plays an important role in the aging process but the effect of age on the gene expression of ANTIOXIDANT ENZYMES has not been extensively studied and findings are contradictory. In this study rats were divided into two groups, Young group (9 months, n = 7) and old group (24 months, n = 7). Animals were sacrificed under deep anesthesia (Ketamine, 100 mg/kg, I.P). Brain of animals were dissected out and cleaned of connective tissues. Brain tissues were immersed inRNALater (Ambion, Austin, TX) overnight at 4°C before storage at –20°C until analysis. RNA extraction was performed using the RNeasy Plus Mini Kit (QIAGEN) following the manufacturer’s protocol and after cDNA synthesis. We compared the levels of expression three genes encoding the ANTIOXIDANT ENZYMES in young and old rat brain, using semiquantitative RT-PCR and analyzing results on a 1% agarose gel. Results showed aging of rats led to an increase of catalase Mrna expression. There was a slight, but significant 20% decrease in MnSOD mRNA during aging. An increase of 40% in Gpx mRNA expression generally occurred during rats aging. This experiment supported a significant difference in gene expression in old rat brain compared to young rat for catalase and superoxide dismutase, and glutathione peroxidase genes.

Abstract: 

Introduction: There is a complex interaction between the hypothalamus, pituitary, and ovary in the human menstrual cycle. There is increasing evidence about the role of oxidative stress on female reproductive tract. Methods: The purpose of this study was to determine the activities of ANTIOXIDANT ENZYMES; superoxide dismutase, glutathione peroxidase, catalase and total ANTIOXIDANT capacity during the menses, follicular and luteal phases of the menstrual cycle in twenty women. In addition, the relationship between the activity of ANTIOXIDANT enzyme and estradiol, progesterone, LH, FSH, and testosterone were evaluated. Results: There was no significant difference between the activity of superoxide dismutase, glutathione peroxidase, catalase and total ANTIOXIDANT capacity during the menses, follicular and luteal phases of the menstrual cycle. We found a significant correlation between LH and FSH with the activity of superoxide dismutase and glutathione peroxidase during the luteal phase (p<0. 05). Conclusion: Our results showed that there were not significant differences in activity of ANTIOXIDANT ENZYMES during the menstrual cycle.

Abstract: 

UNDER DROUGHT CONDITIONS, PLANTS APPLY DIVERSE MECHANISMS AGAINST STRESS. IN THIS STUDY, VARIATIONS&NBSP; OF ANTIOXIDANT ENZYMES ACTIVITIES WERE MEASURED IN LEAVES OF LICORICE PLANTS (GLUCYRRHIZA GLABRA) GROWING UNDER NORMAL AND DROUGHT CONDITIONS. SAMPLES WERE HARVESTED AT 2, 16, 26 AND 28 DAYS AFTER DROUGHT IMPOSITION (S1, S2, S3 AND S4) FROM 10-MONTH-OLD PLANTS. THESE SAMPLES ASSIGNED TO THREE DROUGHT LEVELS, WITHOUT DROUGHT STRESS (S1), MODERATE STRESS (S2) AND SEVERE STRESS (S3 AND S4) BASED ON RELATIVE WATER CONTENT DATA. IN COMPARISON WITH CONTROL SAMPLES, ASCORBATE PEROXIDASE (APX), SUPEROXIDE DISMUTASE (SOD), AND PEROXIDASE (POD) ACTIVITIES WERE UP-REGULATED BY MODERATE AND SEVERE DROUGHT STRESS IN LEAVES. THE MAXIMUM ACTIVITIES OF THESE ENZYMES WERE DETECTED IN SEVERE STRESS. CATALASE (CAT) ACTIVITIES WERE DECREASED IN ALL SAMPLES. THESE FINDINGS SUGGESTED THAT THE HIGHEST ACTIVITY OF SOD, APX AND POD WERE INDUCED BY SEVERE STRESS IN LEAVES.
